A New Era for Aplos NCA: A More Connected, Flexible, and Traceable PK Analysis Workflow

At Aplos Analytics, our goal has always been to make pharmacokinetic analysis faster, more accessible, and more transparent. Over the past year, we have worked closely with customers, trial users, and industry experts to understand how scientists use Aplos NCA in real-world environments and where we could make the biggest impact.

The result is the most significant update to Aplos NCA since its launch.

Coming soon, users will gain access to a completely redesigned platform experience that connects every stage of the workflow, from data preparation and PK analysis to custom calculations, reporting, validation, and archiving.

While the science behind the calculations remains unchanged, the way users interact with and manage their analyses has been fundamentally improved.

A New User Experience Built Around Scientific Workflows

The updated platform introduces a new home page that serves as a central hub for all activities. Users can quickly access data files, analyses, reports, validation tools, documentation, support resources, and administrative functions from a single location.

The redesign was driven by a simple principle: scientists should be able to focus on their analysis rather than navigating software.

The new interface provides a clearer workflow, better organization of projects, and improved visibility across every stage of an analysis.

Flexible Data Cleaning with Full Traceability

Data preparation is often one of the most time consuming steps in pharmacokinetic analysis.

The new Aplos NCA workflow introduces a dedicated data cleaning environment that allows users to build and reuse processing rules directly within the platform.

Users can:

  • Sort and organize datasets
  • Trim whitespace and standardize text fields
  • Perform numeric and unit conversions
  • Add, replace, or modify columns
  • Create iterative cleaning workflows
  • Reload and modify previously applied cleaning rules

Every cleaned dataset maintains a parent-child relationship with its source dataset, creating a transparent audit trail of all transformations applied throughout the workflow.

This structure allows users to revisit previous cleaning steps, make adjustments, and generate updated datasets without starting from scratch.

PK Analysis with Complete Workflow Visibility

Once a dataset is ready, users can move directly into PK analysis.

The new analysis workflow preserves the familiar flexibility of Aplos NCA while improving usability and transparency. Users can configure studies, select parameters, review source data, and execute analyses through a streamlined interface.

Behind the scenes, Aplos NCA maintains a complete hierarchy of all analysis components, including:

  • Original source data
  • Data cleaning workflows
  • Analysis configurations
  • Analysis outputs
  • Supporting files and reports

The result is a fully traceable workflow that makes it easier to understand exactly how results were generated.

More Detailed and Consistent Output Files

The updated platform introduces a standardized output structure designed to simplify downstream workflows.

PK Results files now include both rounded values based on significant figures and maximum precision values generated by the calculation engine. This allows scientists to work with publication-ready values while still retaining access to the full computational precision when needed.

Additional enhancements include improved handling of partial AUC calculations, expanded access to terminal slope estimates, and structured individual profile outputs.

For users who want to investigate alternative terminal slope selections, all KEL estimates remain available within the analysis outputs rather than only the selected best fit result.

Custom Calculations for Study Specific Endpoints

Many studies require calculations beyond standard PK parameters.

The new custom calculation workflow allows users to generate additional endpoints directly from existing PK results.

Examples include:

  • Relative bioavailability calculations
  • Accumulation ratios
  • Exposure ratios
  • Unit conversions
  • Study specific derived PK parameters

Custom calculations become part of the overall analysis hierarchy, maintaining the same traceability and reproducibility as every other workflow stage.

Outputs are standardized, easy to identify, and can be reconfigured or rerun as study requirements evolve.

Reporting and Post Processing Made Easier

The platform now provides a more flexible approach to post processing and report generation.

Users can generate:

  • Individual concentration tables
  • Summary PK parameter tables
  • Spaghetti plots
  • Mean concentration plots
  • Cohort specific visualizations

Reports are built through configurable sections, allowing users to organize outputs in a way that best supports internal reviews, regulatory submissions, or scientific communication.

Because reporting is now integrated directly into the workflow, users can easily generate multiple report versions without recreating analyses.

Advanced Analysis for Power Users

For users who prefer automation and reproducibility, the platform introduces an Advanced Analysis mode.

Instead of moving through each workflow step individually, users can execute an entire analysis using a predefined JSON configuration that includes:

  • Data cleaning
  • PK analysis
  • Custom calculations
  • Tables
  • Plots
  • Reports

This approach enables highly efficient execution of repeat analyses while preserving all workflow documentation and outputs.

Built from Customer Feedback
